Le scénario de Lost Highway se présente comme l'histoire d'un assassin schizophrène racontée du point de vue des différentes personnalités de l'assassin lui-même. Cette perspective inhabituelle est l'une des hypothèses que se forge le spectateur à mesure que l'intrigue progresse. L'histoire est d'abord celle d'un mari jaloux qui assassine sa femme. Plus tard vont apparaître d'autres personnages, aux identifiés parallèles, en réseau avec le premier. Lost Highway est un film noir d'horreur du XXIe siècle, un monde où le temps est dangereusement hors de contrôle, un terrifiant voyage sur la route perdue...
